N = number of cells (sperm)
Area = 9 mm2
Average of cells = mean of cells on both sides of the chamber / 9 squares
Dilution = usually undiluted “0”
Depth = 0.1mm

Concentration cells/ml = average of cells x dilution x 1000(where is this from) / # squares

It comes from the product of area, depth and dillution.
Area = 4
Depth = 1/10
Dillution = 1/20
1/20 × 1/10 × 4 = 4/200 =1/50
Considering that you are dividing cell count by 1/50 thats is equal to multiplying cell count by 50.

One colleague who used Nageotte hemocytometer wondered my decision, since he emphysized his support from Lutz publication at Transfusion Vol 33 No 5 Pages 409-412, 1993. He mentioned Nageotte calculates WBC in 50 uL, however, Neubauer counts WBC only in 0.1 uL x 5. Sampling bias is his concern. Could you please help me solving this question? Thank you very much!

Let N be the no.of wbcs in all the 4 sqaures.
Volume of one wbc square =1×1×0.1=0.1mm3
Volume of 4 wbc squares =0.4mm3
0.4mm3 of wbc squares contain wbcs=N
1mm3 of wbc squares contain wbcs=N/0.4
Multiplying with dilution factor viz20 =N×20/0.4=N×50.
